Chronic Inflammation May Harm the Brain – Here’s How Science is Fighting Back

A recent feature in New Scientist highlights growing evidence that chronic inflammation can have profound effects on mental health and cognition, linking long-term immune system activation to conditions ranging from depression and anxiety to Alzheimer’s disease.

Research shows that while the immune system’s inflammatory response is essential for fighting infection and repairing tissue, persistent low-grade inflammation can “leak” into the brain, disrupting key processes. Cytokines—signalling proteins that trigger inflammation—can cross a weakened blood-brain barrier, affecting neurotransmitters such as dopamine and serotonin, and overactivating microglia, the brain’s immune cells. Over time, this neuroinflammatory state may contribute to cognitive decline and the formation of amyloid plaques, a hallmark of Alzheimer’s disease.

Large-scale studies support this link. Analysis of 1.5 million people in the UK’s Our Future Health cohort found that individuals with chronic inflammatory conditions—like rheumatoid arthritis, multiple sclerosis, or inflammatory bowel disease—have nearly double the risk of anxiety and depression. Longitudinal studies indicate that elevated inflammatory markers in childhood may predict higher odds of depression or psychosis in adolescence, strengthening the case for a causal relationship.

Researchers are now exploring interventions to protect the brain. Anti-inflammatory drugs, including some originally developed for arthritis, show promise in alleviating depression for people with persistent inflammation. Diet and lifestyle modifications are also critical: adherence to a Mediterranean-style diet rich in fruits, vegetables, nuts, fish, and olive oil has been linked to reduced inflammatory markers and lower risks of depression and dementia. Physical activity, mindfulness, and stress reduction further help dampen chronic inflammation.

Additionally, drugs that mimic the hormone GLP-1, such as semaglutide (marketed as Ozempic and Wegovy), are being studied for their anti-inflammatory effects in the brain, independent of weight loss. Early animal studies suggest these drugs may improve cognitive function and reduce Alzheimer’s-related pathology, offering potential new avenues for treatment.

While scientists are still unraveling the complex relationship between inflammation and the mind, New Scientist emphasizes that both lifestyle choices and emerging medical interventions could help protect cognitive function and mental health in the face of chronic inflammation.
