Stem Cell Trials Mark ‘Big Leap’ in Parkinson’s Treatment, But Larger Studies Still Needed

With Parkinson’s projected to affect 25 million people globally by 2050, this emerging field of stem-cell-based neurotherapy offers a rare glimmer of hope for a disease that currently has no cure.

In a major development for neurological medicine, two groundbreaking studies published in Nature have shown that stem-cell therapies for Parkinson’s disease are safe and show early signs of effectiveness. The research, carried out independently in the United States, Canada, and Japan, marks a critical step forward in using regenerative medicine to treat the progressive disorder—though experts stress that larger trials are necessary to confirm clinical benefits.

The studies involve transplanting lab-grown dopamine-producing neurons into patients’ brains, aiming to replace the cells lost to Parkinson’s disease. In both trials, the transplanted neurons survived long enough to produce dopamine, a crucial neurotransmitter that diminishes in people with the condition. Some participants even experienced noticeable improvements in tremors and daily function.

“This is a big leap in the field,” said Malin Parmar, a stem-cell biologist at Lund University in Sweden. “These cell products are safe and show signs of cell survival.”

The research published in Nature marks a turning point after decades of mixed results from earlier attempts, including controversial fetal tissue transplants. These latest trials use more advanced techniques, coaxing stem cells into neural progenitor cells that were then injected into a motor-control region of the brain called the putamen.

In the North American trial, led by Viviane Tabar of Memorial Sloan Kettering Cancer Center, 12 people underwent surgery to receive up to 2.7 million cells. Participants showed dopamine activity in the brain and symptom improvement over 18 months. Meanwhile, the Japanese trial, overseen by Jun Takahashi of Kyoto University, used induced pluripotent stem cells (iPSCs) from adult donors and reported similar survival and dopamine production results.

While these early findings are promising, the trials were small—only 19 people across both studies—and primarily designed to test safety. Researchers caution that subjective improvements could be influenced by the placebo effect. Nevertheless, the results pave the way for larger, more definitive studies. Bluerock Therapeutics, which backed the U.S. trial, plans to launch a phase 3 trial this year, including placebo controls via sham surgeries.

Jun Takahashi’s team in Japan is aiming for conditional approval under the country’s fast-track regulatory system, while also addressing ongoing issues like low cell survival rates and the need for immune-suppressing drugs.

As reported in Nature, these advances could eventually transform how Parkinson’s disease is treated, especially if future trials confirm who benefits most and whether using a patient’s own cells can eliminate the need for immunosuppression. Jeanne Loring, a stem-cell researcher at Scripps Research, is already exploring such personalized approaches.
