Largest Protein Study Reveals Clues to Ageing and Brain Diseases

Breakthrough research backed by Bill Gates and Johnson & Johnson aims to transform detection and treatment of Alzheimer’s and Parkinson’s

An unprecedented international study of human proteins is offering new insights into how neurodegenerative diseases like Alzheimer’s and Parkinson’s develop — and how they might one day be detected and treated earlier. The findings, published this week in Nature Medicine and reported by the Financial Times, mark a major advance in efforts to harness large-scale biological data and artificial intelligence to tackle ageing and brain disease.

The research comes from the Global Neurodegeneration Proteomics Consortium (GNPC), an initiative launched in 2023 that combines academic institutions with the backing of pharmaceutical giant Johnson & Johnson and Gates Ventures, the private office of Microsoft co-founder Bill Gates. The consortium has assembled the world’s largest neurodegeneration-focused protein dataset, encompassing 250 million protein measurements and 35,000 biosamples from across the US and Europe.

“This brings us closer than ever to the day when a diagnosis of Alzheimer’s disease stops being a death sentence,” said Gates, who partially funded the project and has long been involved in global health and dementia research.

The study identified specific protein biomarkers linked to genetic variants associated with Alzheimer’s risk, mapped how these proteins change with age, and uncovered distinct patterns of protein abnormalities that correlate with different neurodegenerative conditions. These findings offer new biological clues into how diseases like Alzheimer’s and Parkinson’s begin and progress — a crucial step toward earlier diagnosis and effective intervention.

“The most immediately exciting part is that the patterns of protein abnormality that predict neurodegenerative diseases reveal new insights into the biology of how these conditions develop,” said Charles Marshall, professor of clinical neurology at Queen Mary University of London. “This paves the way for drug discovery work that could ultimately lead to new treatments.”

Simon Lovestone, global head of discovery and translational research at Johnson & Johnson, echoed that sentiment, calling the dataset an “extraordinary resource” that could fundamentally change how neurodegenerative diseases are studied and treated. He added that, after years of slow progress, the field is now accelerating thanks to technological advances and unprecedented access to data.

Neurodegenerative diseases currently affect more than 50 million people globally — a figure projected to double by 2050 due to ageing populations. Alzheimer’s remains the leading cause of dementia, while Parkinson’s is among the fastest-growing neurological conditions worldwide.

Amanda Heslegrave, a principal research fellow at the UK Dementia Research Institute at UCL, emphasized the importance of the dataset’s size and diversity. “This work shows why scale matters when it comes to understanding complex diseases like Alzheimer’s,” she said.

The research also explores the overlapping biology of Alzheimer’s and Parkinson’s, supporting collaborative drug development efforts. Simon Stott, research director at Cure Parkinson’s, noted that these shared biomarkers could be leveraged to design therapies that target both diseases. Cure Parkinson’s is currently partnering with Alzheimer’s Research UK to test drugs that could benefit patients with either condition.

“Biomarkers are crucial,” Stott said. “They help us understand the disease, monitor patients in trials, and see if experimental drugs are working.”

The full dataset — including anonymized clinical data — is being made freely available online to researchers, in an effort to speed up global collaboration and innovation in the field.

As AI-enhanced biological research continues to evolve, experts hope these findings will not only reshape the future of neurodegenerative disease treatment but also offer broader insights into the ageing process itself.
